Operating principle of the OS30p+ Fluorometer for Rapid Detection of Water Stress

Plants use only a part of the absorbed light energy to carry out photosynthesis.

Excess energy can be dissipated as heat, or it can be re-emitted as chlorophyll fluorescence.

These three processes are in competition, so that an increase in one means a decrease in the others.

Anything that affects photosynthesis will affect the fluorescence emission of chlorophyll.

Therefore, chlorophyll fluorescence is a tool to evaluate the photosynthetic state and detect physiological and environmental changes in plants.

Generally, the fluorescence emitted by chlorophyll accounts for between 1 and 3% of the total light absorbed.

The ADC BioScientific OS30p+ Fluorometer is a portable device, optimized for the rapid detection of plant stress in dark-adapted plants.

The OS30p+ Fluorometer for Rapid Water Stress Detection provides values from the FV/FM protocol and the Strasser OJIP protocol.

The maximum photochemical efficiency is obtained and the stress in a dark-adapted state is checked with the OJIP fluorescence induction curve.

The parameters obtained are: Fo, Fm, Fv/Fm, Fv/Fo, O, K, J, I, P, tFm, A, Mo and PI/ABS.

In this equipment, the analysis unit and the console are integrated, making it possible to make measurements with one hand.

The weight of the meter is 900g.

The internal memory allows storage of up to 160,000 data and 32 OJIP graphics.

And this data can be downloaded through a USB port. The device's battery has an autonomy of 8 hours.

Detection method:

Pulse modulated using a PIN photodiode with 700-750 nm band pass filter.

Test duration:

0.1 seconds to 1.5 seconds. The default saturation pulse duration is set at 1.0 second. Software takes a rolling eight point average to determine FO, and FM, making it idea for both algae and land plants.

FO, FM, FV/FM, FV/FO, graphical display of trace.

Strasser OJIP Test

Actinic illumination:

An array of 650 nm red LEDs adjustable up to 6,000 μmols.

Detection method:

Pin photodiode detector with 700-750 nm band pass filter. Variable sampling rate based on test phase.

Test duration:

3 - 300 seconds.

Test parameters measured:

O, t100μs, t300μs (K), t2ms (J), t30ms (I), P, tFM, A (area above the curve), MO (RC/ABS), PIABS (performance index) FO, FM, FV/FM, FV/FO. FO is measured not estimated.

ABS/RC, TRO/RC, DIO/CS, ETO/RC, TRO/ABS, ETO/TRO, ETO/CS, RC/CSO, RC/CSM, S, M, T.

Up to 32 traces can be overlaid on the graphic display screen, and up to 32 traces per file name can be stored. Colors start to repeat on the viewing screen after 16 traces are overlaid. If traces are not stored, thousands of measurement parameter sets can be stored in a single data file for spider graphing.
